Roma will host the first leg at the Stadio Olimpico in February, before returning to Porto on March 6 to complete the tie.
Roma do not have the best historic record against Porto - losing both European ties against them, in 1981 and 2016 - but Di Francesco is excited at the opportunity to redress the balance.
“Being back in the knockout phase of the Champions League is yet another exciting moment for this club,” said Di Francesco.
“The draw in Nyon has decided we will face Porto, who won more points than anyone else during the group stage and currently sit top of their domestic league.
“I know that the past meetings between these two clubs do not make for great reading for us, but over the course of these two games we will try to turn around that record.”
